OPTIMASI PROSES EKSTRAKSI BUNGA SEPATU (Hibiscus rosa sinensis) MENGGUNAKAN ULTRASONIC-ASSISTED EXTRACTION DENGAN PARAMETER KADAR ANTOSIANIN DAN AKTIVITAS ANTIOKSIDAN
Hibiscus flower contains anthocyanin secondary metabolites which have pharmacological effects as antioxidants. The purpose of this study was to determine the optimal temperature, time, and concentration of the extraction solvent based on the determination of yield, total anthocyanin content and IC50 antioxidant activity.The research was conducted by varying the temperature (25, 42.5, 60), time (10, 20, 30), and solvent concentration (40, 60, 80%). Determination of the best extraction conditions was carried out using a central composite design (CCD). Based on Design Expert 12® analysis, the best ultrasonic extraction conditions were obtained at a temperature of 36oC, a time of 25 minutes, and an ethanol concentration of 80% which was selected based on the highest desirability value. Ultrasonic extraction using a temperature of 36oC, a time of 25 minutes, and 80% ethanol resulted in a yield of 26.4%, anthocyanin content of 419.76 mg/100g and an antioxidant IC50 value of 39.4241 ppm. Based on the characterization test of the extract under optimum conditions which included moisture content, drying shrinkage, and total ash content, the results were obtained that met the requirements of the Indonesian Herbal Pharmacopoeia.
