OPTIMASI DAN KARAKTERISASI TRANSETOSOM FRAKSI ETIL ASETAT EKSTRAK KULIT BUAH MANGGIS (GARCINIA MANGOSTANA L.) DENGAN VARIASI KONSENTRASI TWEEN 80 DAN ETANOL 96%
The mangosteen rind (Garcinia mangostana L.) are proven contains a xanthone-derived bioactive compound in the form of α-mangostin which has ability as an anti-tyrosinase. The ability of transethosomes to penetrate through the stratum corneum can increase the pharmacokinetic value and pharmacological effects of the fractions used. This study aims to determine the optimum formula of varying concentrations of tween 80 and ethanol 96% for the transethosomal characterization of the ethyl acetate fraction of mangosteen rind. Optimization was carried out using the 22 factorial design method using Design-Expert 12® which tween 80 and ethanol 96% as the factors on the response of the encapsulation efficiency value, and encapsulation efficiency reduction and pH reduction after the stability test. The optimum formula was characterized includes particle size, PDI, and zeta potential. The total xanthone content of the ethyl acetate fraction of mangosteen rind was 362.11 mg AME/g extract. Preparation of transethosomal suspension using thin layer hydration method with concentration ratio of tween 80 and ethanol 96% of 0.75:30; 0.4:30; 0.4:35; and 0.75:35. The optimum formula was obtained using 0.75% tween 80 and 35% ethanol 96% which given encapsulation efficiency value of 95.77 ± 0.01, encapsulation efficiency reduction of 2.454 ± 0.008, and pH reduction of 0.117 ± 0.005. The results of particle characterization of the optimum formula obtained particle size of 269,8 nm, PDI 0,457, and zeta potential of -18.9 mV
