Nanoemulsion is a stable drug delivery system. The preparation of nanoemulsion of basil essential oil was carried out to increase the stability and bioavailability of basil essential oil and to increase its antibacterial effect. This study aims to compare the 3% basil essential oil nanoemulsion and 3% basil leaf essential oil which was not formulated in nanoemulsion on the activity of Staphylococcus aureus. In this study, there were 4 formulas produced by the Design Expert 13® using the factorial design method with a tween 80 factor and propylene glycol and their effect was determined on the response to pH, specific gravity, particle size, polydispersity index, and percent transmittance. The optimum formula of basil essential oil nanoemulsion obtained was carried out by centrifugation test, cycling test, antibacterial activity test by disc diffusion method, and determination of MIC and MBC values. From the test, it can be found that the optimum formula of basil essential oil nanoemulsion at a concentration of tween 80 and propylene glycol were 20%. The results of centrifugation showed that there was no phase separation. The results of the cycling test showed an increase in pH from 4.55±0.01 to 4.69±0.006. The value of the inhibition zone diameter of 3% basil essential oil nanoemulsion was 18.33 ± 0.66 and 3% basil essential oil was 11.44 ± 3.68. The MIC values of basil essential oil nanoemulsion and basil leaf essential oil were respectively at concentrations of 62.50 ppm and 250 ppm. The MBC values were at concentrations of 125 ppm and 500 ppm. Based on the results of the study, it can be concluded that the optimum formula of basil essential oil nanoemulsion has strong antibacterial activity compared to basil essential oil which is not formulated in nanoemulsion and it has good stability.
