Cempedak seeds are one of the plants that have the potential to be a source of antioxidants as an antidote to free radicals. Cempedak seeds in ethanol extract contain secondary metabolite compounds such as flavonoid saponins, tannins, steroids where these compounds are thought to be efficacious as hepatoprotectors. This study aims to determine the activity of ethanol extract of cempedak seeds as a hepatoprotector by measuring the levels of SGPT and SGOT as well as histopathological and macroscopic features induced by CCl4. The rats were divided into six groups, namely the normal group (Na CMC 1%), the positive group (Silymarin 200 mg/kgBB), the negative group (CCl4 0.1 mL/kgBB) and the cempedak seed ethanol extract treatment group with a dose of 250, 500, 1000 mg/kgBB. Variations in doses of ethanol extract of cempedak seeds had an effect on decreasing levels of SGPT and SGOT in male white rats of wistar strain induced by CCl4. The larger the dose of food, the greater the effect of decreasing levels of SGPT and SGOT levels. Based on the results of the post hoc analysis of the positive group with test III there was no significant difference. The results of the histopathology of the liver in the normal, positive, negative groups, test I, test II and test III in the form of necrosis conditions 0, 1, 3, 2, 1, 1. Measurement of macroscopic images of liver organs in test group III in accordance with the criteria for normal liver. Based on the relationship between the percentage of hepatoprotector effect on the dose, the ED50 value of the ethanolic extract of cempedak fruit seeds was 230.17 mg/kgBB.
