OPTIMASI PRODUKSI METABOLIT SEKUNDER FUNGI ENDOFIT Neopestalotiopsis sp. DENGAN VARIASI SUMBER C, N, DAN pH
Endophytic fungi that isolated from Bengkal (Neuclea orientalis L.) have the strongest potential for antibacterial activity, which one is Neopestalotiopsis sp. At concentration of 1000 µg/mL, the endophytic fungi Neopestalotiopsis sp. has a strong antibacterial activity of 84,6% against Eschericia coli ATCC8739 and antibacterial activity of 92,8% against Staphylococcus aureus ATCC6538. Endophytic fungi Neopestalotiopsis sp. only produce secondary metabolites of 0,26 g and containts bioactive compounds just only tannins. Based on that, it is necessary to optimize the composition of cultivation media with various sources of carbon, nitrogen, and pH. This research was conducted from August 23, 2021 till December 10, 2021 at the Microbiology Laboratory and the Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The stage of this research consisted of propagation of endophytic fungi, selection the sources of carbon, nitrogen, and pH in cultivation media, extraction of secondary metabolites, optimization of the composition of cultivation media using RSM, extraction of secondary metabolite, and analysis of secondary metabolite with TLC from result of optimization. The formulation for optimization of the medium composition for endophytic fungi Neopestalotiopsis sp. is using glucose 4,179 g L -1 , pepton sebesar 0,449 g L -1 , dan pH 6,954 which is indicated by the optimum secondary metabolite response 0,326 g and desirability value 0,786. Chromatogram profile from the extract of secondary metabolite Neopestalotiopsis sp. Optimization result show of tannins, terpenoids, phenols, and steroids.
