Kersen plant (Muntingia calabura L. (Linn)) has been widely used by the community for natural medicine. Cherry leaves contain flavonoid compounds, tannins, saponins, steroids and triterpenoids that can be used as antibacterial, antioxidant, anti-inflammatory, and antidiarrheal. Salmonella typhi is one of the pathogenic bacteria that causes digestive tract infections and causes diarrhea. The aim of this research was to evaluate the ethanol fraction of cherry leaves as antidiarrheal by using an in vivo of wistar male white rats induced by Salmonella typhi. The treatment group was divided into six groups: negative control, positive control and test group with doses of 30, 60, 90, and 120 mg/kgBW. The negative control group was given NaCMC and the positive control group was given chloramphenicol. Parameters of the initial time of diarrhea in rats was 7 days after rats were induced by Salmonella typhi. Parameters of stool weight, stool consistency, frequency of diarrhea, changes in body weight, number of colonies of Salmonella typhi, percent antidiarrheal effect, percent change in body weight of rats in the test group at a dose of 120 mg/kgBW had an effect close to the positive control group. The results of the characterization of the ethanol fraction of cherry leaves obtained water content of 4.67%, drying shrinkage 5.27%, total ash content of 1.67%, acid insoluble ash content of 0.6%, water soluble extract content of 6.67%, starch content ethanol soluble 33.3%, microbial contamination test 0 cfu/g, and 0.068 mg/kg metal contamination test. The total tannin content in the ethanol fraction of cherry leaves was 174,365 mgTAE/g or 17.43%. Cherry leaves have the potential to be developed into antidiarrheal drugs. The effective dose (ED50) of cherry leaf ethanol fraction as antidiarrheal was 63,548 mg/kgBW. Keywords: cherry leaf, antidiarrheal, fraction¬ ethanol, Salmonella typhi
