Silver nanoparticles have been widely used because of their antibacterial activities. However, the synthesis of silver nanoparticles still uses physical and chemical methods that are not eco-friendly. Therefore, green synthesis of silver nanoparticles was carried out by utilizing the ethanol extract of unripe areca nut which is more eco-friendly. The unripe areca nut contains high levels of flavonoids and polyphenols which can be a bioreduction and stabilizer. This study aims to optimize the silver nanoparticles formula using the simplex lattice design method through the Design-Expert 12® device with various concentrations of silver nitrate (AgNO3) and extract. The study used 9 formulas to determine the maximum wavelength and absorbance values of silver nanoparticles SPR, then characterization of the resulting optimum formula were carried out. The results of the phytochemical screening of the ethanol extract of unripe areca nut contain flavonoids, alkaloids, polyphenols, tannins, and saponins which can reduce and stabilize silver nanoparticles. The flavonoid content in the extract was 110,849 mg CE/g extract. Optimum formula optimization results were obtained from the concentration of silver nitrate (AgNO3) 3 mM and 3 mL of 10% w/v extract. The results from the optimum formula characterization show the maximum wavelength of SPR in 420 nm and absorbance 1.427, the value of particle diameter is 122.53 nm; PDI 0.35; and zeta potential -19.75 mV. The results of the physical stability test showed the formation of irreversible deposits in the 1st to 6th cycles, a shift in the SPR wavelength from 426 nm with an absorbance of 1,385 to 429 nm with an absorbance of 1,246 in the 6th cycle, and a decrease in the pH value from 9,00 to 7.97 in the 6th cycle. Based on the data obtained, the optimum formula has good particle characteristics but lacks physical stability.
