Silver nanoparticles have antimicrobial activity that can inhibit the bacteria and fungi’s growth. An eco-friendly, fast, cost-effective and biocompatible green synthesis method for the synthesis of silver nanoparticles using ethanolic extract of unripe areca nut as a bioreduction and stabilizer. This study aims to obtain the optimum temperature and pH conditions in the synthesis of silver nanoparticles. The ethanolic extract of unripe areca nut was carried out for phytochemical screening and determination of total flavonoid content. In this study, there were 9 formulas of silver nanoparticles produced by Design-Expert 12® using response surface methodology and λmax surface plasmon resonance (SPR) response and absorbance were determined and analyzed to produce the optimum formula. The optimum formula for silver nanoparticles was characterized and stability tested. The results of phytochemical screening showed that the ethanolic extract of unripe areca nut contains alkaloids, polyphenols, tannins, flavonoids and saponins. The total flavonoid content in the ethanolic extract of unripe areca nut was 110,8 ± 0,4110 mgCE/g extract. The optimum formula for the synthesis of silver nanoparticles using ethanolic extract of unripe areca nut was obtained at temperature of 30°C and pH of 10,5. The characterization results of the optimum formula showed the λmax SPR value of 423 nm, absorbance 1,148, particle size of 161,7 ± 46,1 nm, PDI of 0,286 ± 0,035, zeta potential of -16,1 ± 3,7 mV. The stability test showed the formation of precipitate in the 1st to 6th cycle, a redshift in the value of λmax SPR from 423 nm with an absorbance of 1,148 to 425 nm with an absorbance of 1,396 in the 6th cycle and a decrease in pH from pH 10,50 ± 0,008 to pH 10,10 ± 0,008 in the 6th cycle. The optimum formula has good particle characteristics, but it still has less stability.
