Cempedak is an endemic plant of Indonesia. Cempedak leaves (Artocarpus integer (Thunb.) Merr.) contain secondary metabolites such as flavonoids, steroids, tannins and saponins which have analgesic and anti-inflammatory activity to inhibit the formation of prostaglandins. This study aimed to determine the characteristics of the extract also its analgesic and anti-inflammatory effects based on the decreasing in rat writhing induced by acetic acid 0.6% v/v and based edema volume induced by carrageenan 1% w/v. Analgesic and anti-inflammatory tests were carried out using the writhing test and rat paw edema method on 24 Wistar male white rats which were divided into 6 groups, namely the positive control group which was given Diclofenac Sodium, the negative control group which was given 1% Na CMC, and the test group (ethanol extract), n-hexane fraction, ethyl acetate fraction and water ethanol fraction) at a dose of 200 mg/kgBW. Parameters observed included the number of writhing, %analgesic, volume of rat edema, %inflammation and %inflammation inhibition. The percentage value of the analgesic effect of Diclofenac Sodium was 80,42%, ethanol extract was 65,74%, ethyl acetate fraction was 76,75%, n-hexane fraction was 47,15% and ethanol-water fraction was 34,86%. The %inflammation inhibition of Diclofenac Sodium was 61,49%, ethanol extract 53,53%, n-hexane fraction 45,90%, ethyl acetate fraction 55,84% and ethanol-water fraction 35,76%. Data on the number of writhing, percent of analgesic, percent of inflammation and percent of inhibition were analyzed using One-way ANOVA to see differences between groups. The results showed that the characterization of the ethanol extract of cempedak leaves met the requirements of the Ministry of Health RI 2008. The extract and fraction of cempedak leaves had analgesic and anti-inflammatory activity. The ethyl acetate fraction gave the greatest analgesic and anti-inflammatory effect compared to ethanol extract, n-hexane fraction and water-ethanol fraction and did not differ significantly (p>0.05) with Diclofenac Sodium.
