One of the Indonesian plants that has the potential as an antioxidant is breadfruit (Artocarpus altilis [Park.] Fosberg). The leaves contain flavonoids which have strong antioxidant power. This study was conducted to determine the total flavonoid content of the ethyl acetate fraction of breadfruit leaves, the effect of the preparation of the ethyl acetate fraction of breadfruit leaves on MDA levels, catalase enzyme activity, macroscopic and histopathological descriptions of male white rats of Wistar strain induced by CCl4 and to determine the best dose as an antioxidant. This study used 30 rats which were divided into 6 groups, namely normal control and negative control (Na-CMC 1%), positive control (vitamin C 10 mg/kgBW), and the test group at a dose of 25 mg/kgBW, 50 mg/kgBW, and 100 mg/kgBW. The results showed that the total flavonoid content of the ethyl acetate fraction of breadfruit leaves was 29,41 mg QE/g fraction. The three doses of the ethyl acetate fraction of breadfruit leaves had an antioxidant effect and a dose of 100 mg/kgBW had the best effect because there was no significant difference with the positive control (P>0,05). The conclusion of this study is that the variation in the concentration of the ethyl acetate fraction of breadfruit leaves affects MDA levels, catalase enzyme activity, macroscopic and histopathological features in rats with a dose of 100 mg/kgBW, the ethyl acetate fraction of breadfruit leaves can reduce plasma MDA levels and increase liver catalase enzyme activity and provide good macroscopic and histopathological picture of the liver.
