Binjai (Mangifera caesia Jack.) is a plant that is empirically used by the people of Bangka Belitung as an alternative treatment for pain and inflammation. Binjai leaves contain secondary metabolites such as flavonoids, tannins, and steroids that have the potential as analgesic and antiinflammatory. This study aims to determine the characteristics of the extract, the effect of extract and fraction of Binjai leaves as analgesic and antiinflammatory, and the compounds in the active fraction of Binjai leaves. The Wistar rats were divided into 6 groups, namely the negative control group, the positive control group which was given 4.5 mg/KgBW of Diclofenac Sodium, the ethanol extract, the n-hexane fraction, the ethyl acetate fraction and the ethanol-water fraction at a dose of 300 mg/KgBW. Pain was created by inducing acetic acid 0.6% w/v intraperitoneally one hour after administration of the solution samples. Inflammation was created by inducing carrageenan 1% w/v subplantar one hour after administration of the solution samples. Parameters observed included the amount of writhing, %analgesic, %inflammation, and %inflammation inhibition. The data were analyzed using One Way ANOVA test to see the difference in the percentage of analgesic and antiinflammatory effects between groups. The characterization results showed that the ethanol extract of Binjai leaves met the parameter limits according to the Indonesian Ministry of Health (2008). The test results showed that the ethyl acetate fraction gave the largest percentage with analgesic percentage was 74.02% and inflammation inhibition percentage was 60.11% followed by ethanol extract, n-hexane fraction, and ethanol-water fraction.
