Kenikir leaves (Cosmos caudatus Kunth.) is a plant that contains many active compounds, such as flavonoids, saponins, and tannins that have anti-inflammatory, antioxidant, antibacterial potential and also accelerated the process of open wound healing. This study aims to determine the variations in the concentration of ethanolic extract of kenikir leaves that affect the characterization of spray gel preparations and affect the healing activity of open wounds. Variations in the concentration of extracts used in this study were 10%, 15%, 20%, and 25%. Spray gel preparation qualiy were analyzed which included organoleptic, homogeneity, viscosity, pH, adhesion, spraying patterns, pump delivery, washing ability and stability tests. Wound healing activity testing was carried out with 8 treatments, contained 4 rats for each group. It consisted of a positive control with Tekasol®, negative control, treatment group with concentration of 10%, 15%, 20%, 25%, 15% extract group and placebo. The activity test of the spray gel preparation on open wounds was carried out by presenting the gel preparation as much as one spray every 24 hours for 20 days. The test of the activity of the spray gel preparation against open wounds is carried out by observing the diameter of the wound on the 4th, 8th, 12th, 16th, 20th day and %recovery calculation. The data were analyzed using One Way ANOVA test to see the difference in the percentage of exhalation (%recovery) between groups. The results of the preparation characterization showed that all spray gel formulas fulfilled the requirements of the physical evaluation test of the spray gel. The results of the activity test showed that spray gels with extract concentrations of 25% gave the best %recovery was 100.00 ± 0.00 with the fastest healing time for 12 days. As for the preparation with an extract concentration of 10%, the preparation with the lowest %recovery was 97.613% for 21 days. Statistical analysis showed that the administration of spray gel preparations of ethanolic extract of kenikir leaves could accelerate the healing of open wounds, this was indicated by a significant difference (p
