OPTIMASI FORMULA SERUM ANTIAGING EKSTRAK ETANOL DAUN KOPI ROBUSTA (Coffea canephora) DENGAN VARIASI CARBOPOL®934 – HPMC –KITOSAN MENGGUNAKAN CENTRAL COMPOSITE DESIGN
Robusta coffee leaves (Coffea canephora) contain phenolic compounds, one of which is chlorogenic acid, which has the potential as an antioxidant that can counteract free radicals that cause premature aging. The lack of use of robusta coffee leaves as natural ingredients that are efficacious in the development of antiaging preparations is expected to be able to make a serum formula for ethanol extract of robusta coffee leaves as antiaging preparations. This study aims to obtain the optimum conditions of the serum formula of robusta coffee leaf ethanol extract. Robusta coffee leaves were extracted using the Ultrasound-Assisted Extraction method and yielded a yield of 13,90%. The total phenolic content in the ethanolic extract of robusta coffee leaves was 50,32 mgGAE/g extract. Robusta coffee leaf ethanol extract was formulated into a serum dosage form using various gelling agents carbopol®934, HPMC, and chitosan. The three gelling agents were optimized using the Design Expert 12 program with the Central Composite Design approach which obtained 15 run formulas. The preparations were evaluated for physical properties, namely organoleptic, homogeneity, adhesion, pH, dispersibility, and viscosity. The test results were analyzed with DX 12 to get the optimum formula. The optimum concentration of carbopol®934 was 0,878%, HPMC 1.135%, and chitosan 0,193% had adhesion of 4,2 seconds, spreadability of 5,1 cm, and viscosity of 876,641 cP.
