PREPARASI DAN KARAKTERISASI NANOCARRIER KITOSAN-ALGINAT PEMBAWA ASAM USNAT DENGAN VARIASI KONSENTRASI CaCl2 MENGGUNAKAN METODE GELASI IONIK
The potential utilization of usnic acid is still very limited due to its low solubility in water causes low bioavailability, so that its therapeutic effectiveness also decreases. Chitosan-Alginate polymer based nanocarrier formulation uses ionic gelation method as a new delivery system, so that it would facilitate the absorption process and increase the effectiveness of the usnic acid to achieve the target of treatment. This research aims to obtain the best nanocarrier formula that has good characterization. This research characterizes usnic acid isolate. The study used 3 formulas with various concentration of CaCl2 20 µL, 40 µL, and 100 µL. The optimum formula will be determined based on the highest %EE value and also carried out particle characterization and stability test. The result of the characterization of usnic acid isolate obtained wavelengths of 287 nm in water medium and 230 nm in methanol medium, melting point 204,3-204,8oC, and TLC with Rf 0,42. The first, second, and third formulas had %EE respectively of 95,89%; 96,43%; 98,49%. The characterization results showed the particle size value of 246,3 nm; PDI 0,412; and zeta potential +26,93 mV. Stability test showed that usnic acid nanocarrier in three storage conditions with release model following zero orde, activation energy 6,28 cal/mol, kinetic constant 0,083 mg/days with shelf-life 59 days at temperature 25oC and estimated shelf-life 71 days at temperature 5oC.
