Glibenclamide is one of the drugs classified to class II in the BCS (Biopharmaceutics Classification System), which means this substance has low solubility in water but has high permeability. The manufacture of solid lipid nanoparticles using stearic acid, PVA, Tween 80 and PEG 1000 aims to increase the solubility of glibenclamide in the body using high homogenization and ultrasonication methods. This study aims to determine the effect of variations in the concentration of cosurfactant PEG (Polyethylene Glycol) 1000 in the formulation. The formulas in this study were varied into 3 formulas with a variation of PEG 1000 as much as 0.25%; 0.5%; 1% and the best formula was obtained based on the value of encapsulation efficiency (%EE) using a UVVisible spectrophotometer and particle characterization was carried out using a Particle Size Analyzer, stability test and solubility test. The best formula was obtained in formula 1 which had a percent EE value was 87.8303±0.03 and pH was 6.31. The percentage of EE for formula 2 was 84.0205±0.02 and formula 3 was 83.1038±0.03. The characterization results showed the particle size of solid lipid nanoparticle was 443±20.08 nm, a polydispersity index value 0.489 and a zeta potential was -14.3±3.82 mV. The results of the solubility test showed that the solid lipid nanoparticles were soluble in aquadest, medium with acidic pH, and SIF (Simulated Intestinal Fluid) medium. The results of the physical test using the heating-cooling method of solid lipid nanoparticles showed a decrease in pH from 6.31 to 4.42 and a white precipitate was formed. Mechanical test by centrifugation showed a surface formed. The results obtained, formula 1 (0.25% PEG1000) is the best formula but did not have a good stability.
