DNA BARCODING HORSESHOE CRAB ASAL PERAIRAN KUALA TUNGKAL, JAMBI DAN PESISIR BANYUASIN, SUMATERA SELATAN
Conservation is one solution to preserve the horseshoe crab, which has been categorized as an endangered biota. With DNA barcode, horseshoe crab can be conserved based on its species. This study aims to identify and determine the relationship between horseshoe crab by using DNA barcode. This research carried out from July to November 2021 in the Kuala Tungkal Waters, Jambi and Banyuasin Coastal, South Sumatera with 13 station points by taking 23 horseshoe crabs. In situ measurements consist of temperature, salinity, pH, DO and depth. Sample preparation was carried out by extracting genomic DNA using the GeneAid Kit, DNA amplification using the PCR Thermal Cycler and DNA visualization using Electrophoresis. The data were analyzed with BioEdit and MEGA 11, then identified with the NCBI website. The identification result using DNA barcode in Banyuasin Coastal, South Sumatera consisted of C. rotundicauda and T. gigas species, while in Kuala Tungkal Waters, Jambi consisted of C. rotundicauda species. Based on the phylogenetic tree, it can be seen that there is a relationship between C. rotundicauda at the two research sites, which indicates the occurrence of gene from one place to another. The origin of genetic diversity of the T. gigas horseshoe crab in Banyuasin comes from Singapore, C. rotundicauda in Banyuasin comes from Johor, Korea and China. Meanwhile, C. rotundicauda in Kuala Tungkal came from Johor and Korea.
