The ethanolic extract of Mangkokan leaf (Polyscias scutellaria (Burm.f.) Fosberg.) contains flavonoids, alkaloids, phenolic, saponins, and steroids that are potentially active as analgesics and anti-inflammatory with their ability to inhibit the formation of prostaglandins the main mediators of pain and inflammation. This study aims to determine the characteristics of the extract and to evaluate analgesic and anti-inflammatory effects of the ethanol extract and the fraction with different polarity of the solvent of the Mangkokkan leaf based on decrease in the writhe from acetic acid 0.6%v/v and decrease volume of edema from carrageenan 1%w/v induction. Analgesic and anti-inflammatory study was carried out by using writhing test and rat paw edema methods in 24 male Wistar rats which were divided into 6 treatment groups. Group I as a negative control was given 1% Sodium CMC, group II as a positive control was given diclofenac sodium, and groups III, IV, V, and VI as a test group with the same dose 400 mg/kgBW and differences in solvent polarity (ethanol extract, n-hexane, ethyl acetate, and ethanol-air fraction). The results of the characterization of the extract met the requirements of the Depkes RI in 2008. The percentage values of analgesics and anti-inflammatory for positive controls were 80.42% and 61.49%, treatment groups III;IV;V; and VI respectively were 77.68% and 60,75%; 76.45% and 49.32%; 69.42% and 39.81%; and 64.53% and 28.39%. This indicates that the ethyl fraction is the most active and optimal as an analgesic and anti-inflammatory and the resulting effect is equivalent to the positive control treatment with diclofenac sodium (p>0.05).
