The waste is a habitat for fecal bacteria (coliform), and it is suspected to contain various strains of coliform bacterial phages. Escherichia coli is usually used as an indicator to see the water quality. Therefore, to avoid contamination, biocontrols such as bacteriophages are needed. Bacteriophages (phages) can be used as an effective solution for the biocontrol of innated pathogenic bacteria because they are natural enemies of bacteria that many found in wastewater.And usually to detect lytic phages in wastewater capable of lysing bacteria. The plaque assay method was used for unit testing of viral infection. When the virus begins its infection in the host cell, it forms what is known as a plaque. This research was conducted from April to November 2021, at the Genetics and Biotechnology Laboratory, at the Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. This method uses the plaque assay test to determine the unit of viral infection. The source of the water samples taken was from the disposal of household and livestock waste in Indralaya as opposite the UNSRI Main Gate, UNSRI Student Dormitory retention pond, Layo Market. Each water sample has a different temperature and pH Based on the research that has been done, it can be seen that the location of the bacteriophage is detected by the formation of plaque on the cup. The characteristics of plaque in each place are different, where each plaque has a different size, and different plaque characters are clear or unclear. Therefore, the results of the isolation of bacteriophages from these sites were successfully isolated against Escherichia coli. The results of the effectiveness of LI 1 with a phage concentration of 7,6 x 107 PFU/mL had the highest effectiveness with a low concentration, PL 1 with a phage concentration of 3,2 x 109 PFU/mL, and LI 2 with a phage concentration of 3,4 x 109 PFU/mL, has the opposite effect. While the PL 1 phage can lyse pathogenic bacteria such as Enterotoxigenic Escherichia coli. And this bacteriophage isolate resulted in damage/splitting of Escherichia coli cells. Meanwhile, the lysis effectiveness test showed that the bacteria inoculated without phage (control) began to show an increase in the number of cells based on the optical density value after 30 minutes of incubation and continued to increase until 8 hours the control itself did not add phages, from the results of the curve, the OD value every hour increases.
