Silver nanoparticles have potential as solution to problem of bacterial resistance because of their antibacterial ability. The green synthesis method using ethanol extract of unripe areca nut was chosen because of its flavonoid and polyphenol content which is able to reduce and stabilize silver nanoparticles. The characteristics of silvernanoparticles influenced by the synthesis conditions, including speed and stirring time. This study aims to optimize the speed and stirring time of the synthesis of silvernanoparticles using ethanol extract of unripe areca nut. Optimization was carried out using the 22 factorial design method through Design-Expert 12® for the maximum wavelength and absorbance of SPR. The optimized formula was characterized by size, PDI, zeta potential and stability test. The results of phytochemical screening of ethanol extract of unripe areca nut showed the presence of flavonoids, alkaloids, polyphenols, tannins, saponins. The total flavonoid content was 110,84 mg CE/g extract. The optimization results obtained a formula with a speed of 1000 rpm for 60 minutes showed the optimum formula which showed a maximum wavelength 419 nm and absorbance 1.515. The results of the characterization of optimized silvernanoparticles showed a particle size of 68.11 ± 4.047 nm, PDI 0.408 ± 0.100, and zeta potential of -9 ± 0.947 mV. The results of the physical stability test were observed that there was no color change, a precipitate was formed starting from the 1st cycle. The maximum wavelength shift occurred from the 0th cycle by 419 nm to 427 nm in the 6th cycle and the decrease in absorbance from 1.515 to 1.365, and decreased in pH from 10.49 ± 0.006 to 9.14 ± 0.010. Based on research, it can be concluded that the stirring speed and time affect the characteristics of silver nanoparticles. The optimum formula silver nanoparticles showed good particle characteristics but lacked stability.
