Kepok banana peel (Musa balbisiana) contains many secondary metabolites, one of which is the alkaloid compunds that has antibacterial activity. The aims of this study to determined the physical stability of the optimum formula for antiacne serum ethyl acetate fraction of kepok banana peels. Kepok banana peel ethanol extract was fractionated by the liquid-liquid. The ethyl acetate fraction of kepok banana peels was formulated into serum preparations using a variety of bases HPMC 60SH and Carbopol 934. Both bases were optimized using the Design Expert 12® program with a Factorial Design approach which obtained 4 formulas. Serum preparations were evaluated for physical properties including organoleptic, homogeneity, pH, dispersibility, adhesion and viscosity. Based on the analysis of Design Expert 12®. The optimum formula obtained was tested for stability and antibacterial activity against P. acnes bacteria. The ethyl acetate fraction with a yield of 67.85% indicated the presence of flavonoid compounds, alkaloids, polyphenols, tannins and saponins. Based on the analysis of Design Expert 12®, the optimum formula was obtained with a concentration of HPMC 60SH 0.75% and Carbopol 934 0.75%. Physical stability of the optimum formula for serum based on the stability test of the centrifugation method at 3800 rpm did not occur phase separation, while the cycling test showed an increase in pH from 4.88 ± 0.02 to 5.150 ± 0.035. The antibacterial activity test of the optimum serum formulation against P. acnes was carried out using the well diffusion method and the diameter of the inhibition zone was 23.530 ± 1.078 with a very strong category. The optimum formula for the antiacne serum ethyl acetate fraction of the kepok banana peel, had good physical stability and very strong antibacterial activity.
