ISOLASI SENYAWA STEROID DARI TUMBUHAN LUDWIGIA OCTOVALVIS DAN UJI ANTIBAKTERI
Ludwigia octovalvis is one of the plants that often used as traditional medicine. Previous researchers have been reported some bioactivity from the extract or compound isolated from this plant such as antidiabetic, cytotoxic, antioxidant and antibacterial. However, there are no study have been reported about isolated compound and antibacterial activity from the fruits of this plant. Therefore, this research aims to isolate secondary metabolite compound and to test antibacterial activity from isolated compound, methanol extract, ethyl acetate fraction, n-hexane fraction against bacteria Staphylococcus aureus and Shigella dysentriae using disk diffusion method with suspension bacteria 108 CFU/mL. The isolation and purification processes of compound were done by extracting L. octovalvis fruits using a methanol, then liquid-liquid partition using a n-hexane continiued with ethyl acetate. The isolation and purification continued by using VLC (Vacuum Liquid chromatography), eluting crytal by a solvent, and flash column chromatography. From these processes, two white crystalline compounds were obtained, namely compound D (77,7 mg) and compound D6 (2,8mg). Based on elucidation of structure with IR, GC-MS, 1H-NMR, and comparison of 1H-NMR spectra showed compound D was a mixture of three steroid compound, which the major compound was β-sitosterol and the minor compound was campasterol and stigmasterol. Compound D6 was the steroid compound namely ergosterol peroxide. Antibacterial activity of ergosterol peroxide was not indicated activity to against S. aureus and S. dysentriae. Antibacterial activity of methanol extract showed strong activity based on Chan et al. (2007) classification at the highest concentration (60.000 ppm) (9-13 mm) against both of bacteria. Ethyl acetate fraction showed strong activity at 60.000, 30.000 (12-15 mm) against S. aureus, while the strong activity at 60.000 and 30.000 ppm and 15.000 ppm (18-23 mm) against S. dysentriae. n-hexane fraction indicated weak activity at 7.500 ppm (8-9 mm) against S. aureus, but this fraction was not indicated activity against S. dysentriae.
