UJI AKTIVITAS ANTIOKSIDAN ESKTRAK DAUN GANDARUSA (JUSTICIA GENDARUSSA BURM. F.) MENGGUNAKAN METODE DPPH (1,1 DIPHENYL-2 PICRYL HYDRAZIL) DAN FRAP (FERRIC REDUCING ANTIOXIDANT POWER)
Gandarusa (Justicia gendarussa Burm. F.) belongs to medicinal plant that has been used traditionally in the treatment of degenerative diseases. The aims of this study to analysis the total phenolics and flavonoids compounds of the J. gandarussa leaves fraction and its antioxidant activity assay. The research begins with fractionation by maceration method using solvents with increasing polarity (n-hexane, ethyl acetate and methanol). Each fraction were analysis the total phenolics, flavonoids and steroids content using the photometric method and the antioxidant activity were determined using the DPPH (1,1diphenyl-2 picryl hydrazil) and FRAP (Ferric Reducing Antioxidant Power) methods. The results showed the total phenolic content for the fractions of n-hexane, ethyl acetate, and methanol 6.06 ; 8.42 and 13.05 mgGAE/g respectively, while the levels of flavonoids 0.91; 2.64 and 6.56 mgQE/g respectively. The antioxidant activity with the DPPH method showed IC50 values > 1000 mg/L for the all fractions, and was classified as weak antioxidant. The FRAP method showed antioxidant activity with RC50 values for the n-hexane 891.30 mg/L and ethyl acetat 243.21 mg/L (belonging to the moderately active category of antioxidant), and methanol fraction 64.49 mg/L (strong antioxidant category). The antioxidant activity each fractions were relevan of the total phenolic and flavonoid content. J. gendarussa leaves fractions showed strong antioxidant activity in FRAP compared DPPH method.
