UJI AKTIVITAS ANTIBAKTERI EKSTRAK ETANOL DAN FRAKSI ETIL ASETAT DAUN PEPAYA (CARICA PAPAYA) UNTUK PEMBUATAN GEL HAND SANITIZER
Antibacterial activity of ethanol extract and ethyl acetate fractions of papaya leaves for preparations of gel based hand sanitizer had been carried out on this research. Papaya leaves was extracted by maceration method using ethanol. Ethanol extract was fractionated using ethyl acetate. Total alkaloid and flavonoid contents of the extract and fraction were examined using UV-Visible Spectrophotometer. Total alkaloid content in ethanol extract and ethyl acetate fraction were 18.71 and 20.42 QE mg/g extract respectively, while total flavonoid content were 33.86 and 31.44 QEE mg/g extract respectively. Antibacterial activity from the ethanol extract and ethyl acetate fraction were tested using disc diffusion method against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Staphylococcus epidermidis. Ethyl acetate fraction of papaya leaves at concentration of 20 % showed inhibitory diameters of 19.3 mm, 12.3 mm, 12.6 mm and 13.6 mm respectively against four bacteria. Based on the antibacterial activity data, ethyl acetate fraction of papaya leaves at concentration of 20 % has a strong inhibition, so it was used as the active ingredient for hand sanitizer. Antibacterial activity of hand sanitizer gel formulation were tested against four bacteria and showed inhibitory diameters of 13.6 mm, 10 mm, 11.3 mm and 10.6 mm respectively. The characteristic properties of hand sanitizer gel formulation is in accordance with SNI 06-2588-1992 for quality standards of hand sanitizer which resulted in pH 5.53, spreadability 5.7 cm, good organoleptic and homogeneity as well as good physical stability and antibacterial activity for 14 days of storage.
