UJI AKTIVITAS ANTIOKSIDAN EKSTRAK DAUN BUNGA KATARAK (Hippobroma longiflora) MENGGUNAKAN METODE DPPH DAN FRAP
Cataract Flower (Hippobroma longiflora) has used traditionally for cataract treatment which is related to the content of antioxidant compound. H. longiflora leaves was extracted by maceration method using solvents with increasing polarity (n-hexane, ethyl acetate and methanol). The each fraction was analyzed for total fenolic, flavonoid and steroid contents using photometric method. The antioxidant activity was determined using DPPH (1,1- diphenyl-2-picryl hydrazil) and FRAP (Ferric Reducing Antioxidant Power) methods. The result showed that n-hexane, ethyl acetate and methanol fraction of H. longiflora contained fenolic total contents 2.03; 6.79; 10.87 mgGAE/g , the flavonoid contents are 0.90; 5.35; 7.15 mgQE/g and the steroid contents are 154.08; 23.77 and 14.64 mgCE/g respectively. The antioxidant activity analyzed by DPPH method shows that methanol fraction of H. longiflora has IC50 value 731.19 mg/L which means moderate antioxidant acitivity; while n-hexane and ethyl acetate fractions have IC50 > 1000 mg/L which mean it has weak activity. FRAP method shows that all fractions (n-hexane, ethyl acetate and methanol) have RC50 value < 200 mg/L with RC50 values 166.45; 179.06; 57.09 respectively. All fractions (n-hexane, ethyl acetate and methanol) of H. longiflora leaves showed the stronger antioxidant activity with FRAP method rather than with DPPH method. The antioxidant activity is in line with total of fenolic and flavonoid content and the higher activity was showed by methanol fraction.
