ISOLASI SENYAWA METABOLIT SEKUNDER DARI EKSTRAK N-HEKSANA KAYU BATANG BERIANG (PLOIARIUM ALTERNIFOLIUM) DAN UJI AKTIVITAS ANTIOKSIDAN
Beriang plant (P. alternifolium) is a species of the Theaceae family. This plant has not reported yet on the isolation of compounds and antioxidant bioactivity from plant stem wood. This study aims to isolate secondary metabolites, characterize isolated compounds, reveal secondarymetabolites and determine antioxidant activity against n-hexane extract. The purification process for secondary metabolites was carried out by extracting P. alternifolium stem wood with n-hexane as solvent. Purification and separation were continued by using the flash column chromatography (FCC) method and purification the crystals using a methanol solvent. The isolation process produced two compounds, namely compound 1 as much as 4.8 mg and compound 2 as much as 25.9 mg. The isolated compounds were analyzed using UV, 1H-NMR, 13C-NMR, 2D-NMR spectra data and GC-MS. Based on elucidation and analization using GC-MS showed that two isolated compound still had a mixture with other compounds. Compound 1 is a steroid group compound with a stigmastan base framework and has a conjugated carbonyl group, namely the β-sitostenone compound. Meanwhile, compound 2 is a steroid group namely the β-Sitosterol compound. Antioxidant activity against n-hexane extract using various concentrations (50; 25; 12.5; 6.25; 3.125; 1.5625 ppm). The results of the antioxidant activity test on the n-hexane extract showed that the n-hexane extract was not active as an antioxidant with an IC50 value of 234.73 ppm.
