OPTIMASI PRODUKSI METABOLIT SEKUNDER FUNGI ENDOFIT Peniophora sp. DENGAN VARIASI SUMBER C, N, DAN pH
The extract of the secondary metabolites of endophytic fungus Peniophora sp. at concentration 1000µg/mL had a strong antibacterial activity against Escherichia coli ATCC8739 81,81% and against Staphylococcus aureus ATCC6538 78.57%. Endophytic fungus Peniophora sp. produces secondary metabolites such as phenols and alkaloids, but the the secondary metabolites produced was only 0.146 grams, so it is necessary to optimize the production of secondary metabolites through optimization of the composition of cultivation media with variations of carbon sources, nitrogen source, and pH. This research was conducted from August 23, 2021 to December 10, 2021 at the Microbiology Laboratory and the Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The research stages consisted of propagation of endophytic fungus, cultivation with variations of carbon source, nitrogen source, and pH, first extraction, optimization of components of cultivation media with RSM, second extraction, thin layer chromatography analysis, and data analysis. The formulation of the optimization medium for the endophytic fungus Peniophora sp. is using glucose 3.66 g L-1, peptone 0.475 g L-1, and pH 7.034 which is indicated by the optimum secondary metabolite response 0.323 grams and desirability value 0.795. The secondary metabolite extract of the endophytic fungus Peniophora sp. contains phenolic compounds, terpenoids, and flavonoids.
