IDENTIFIKASI POLIMORFISME GEN mTOR rs2536 DAN rs2295080 SERTA EKSPRESI ER, PR, HER-2, Ki-67 PADA PENDERITA KANKER PAYUDARA
Introduction: Dreast cancer is a type of cancer that occurs due io neoplastic transformation of epithelial cells in the breast. This neoplastic transformation is resulted from the accumulation of genetic and epigenetic aberrations. Breast cancer is the most common cancer in the world. One of the genetic factors associated with breast cancer is mTOR gene polymorphism which mostly occurs in 3’ untranslated region and promoter region. mTOR gene polymorphism leads t o constitutional activation of mTOR translation which will initiate signalling network t o nucleus t o increase cell proliferation and differentiation. The aim of this study w as to identify the rs 2536 and rs 2295080 mTOR gene polymorphism with ER, PR, HER-2 and Ki-67 expression protein in breast cancer patients at Mohammad Hoesin General Hospital Palembang. Methods: This study was a descriptive observational study with PCR-RFLP technique using EcoRI and Fokl enzyme on thirty three patients with breast cancer. Results: rs 2536 gene polymorphism in mutant genotype (CC) found in 27 patiens (81,8%). The C allele frequency was 89,4% and the T was 10.6%. The most common genotype found in rs 2295080 gene polymorphism is wild type (TT) fo und in 19 patients (57,6%). The T allele frequency was 63,6% and the G was 36,4%. The data from Anatomical Pathology showed a tendency to increase the expression of Ki-67, ER, PR and HER-2 in rs 2536 and rs 2295080 gene polymorphism. Conclucion: rs 2536 mTOR gene polymorphism (CC) in breast cancer is more common than r s 2295080 gene polymorphism (GG).
