This research is aimed to know the validity and realibility of three-tier diagnostic test of energy flow ecological material feom students of SMA in the Prabumulih City. This study uses the Treagust development methodology, which consists of 10 steps and 3 main stages, namely content definition, obtaining information about students' misconceptions, and developing diagnostic test instruments. The material developed is based on the Campbell book. The three tier diagnostic instrument is made in the form of mutiple choice with CRI. The instrument result is validated by 5 validator that is used with CVR and get the 1 value which means, the three tier diagnostic instrument is valid and relevant in clarifying the misconception. The realibility test of KR-20 is used to test the instrument. From the test result, misconception from each sub material are 45,54% for the food chain, 49,3% for the food nets, 41,2% for the energy flow, 40,5% for the ecology pyramid, 47,5% for the biomassa pyramid. 41% for the total of pyramid, and the 52,1% for the energy of pyramid.
