Plasmodium falciparum causes of severe malaria or cerebral malaria and main cause of the death in malaria case. Malaria still as a health problem in tropic and subtropic countries and also in Indonesia. Malaria caused by Plasmodium falciparum is many appeared in endemic region in Indonesia. In most of region still use Chloroquine as a first line and the combination Sulfadoxine-Pyrimetamine as a second line. Resistance case of P. falciparum to Clhoroquin and Sulfadoxine-Pyrimetamine have been reported in the world included Indonesia. In Africa, Chloroquine use widely cause the resistance fastly and also Sulfadoxine-Pyrimetamine. Main mechanism ofresistance P. falciparum to antimalaria drug is gene mutation. Mutation ofpfcrt and pfmdr gene related to P. falciparum resistance to Chloroquine. Mutation of dhps gene related to P. falciparum resitance to Sulfadoxine and dhfr gene mutation related to P. falciparum resistance to Pyrimetamin. Pfcrt and pfmdr gene mutations are more often appeared than dhps and dhfr gene mutations. The aim of research is identification A437G dhps gene mutation that related to P. falciparum resistance to Sulfadoxine in Sumatera Selatan. the result of research shows that wild-type (normal) genotype A437G dhps gene was 100% (30 from 30 isolate). Mutation A437G of dhps gene related to Sulfadoxine resistance of malaria patient infected by P. falciparum in South Sumtatera during MarchSeptember is not occurred yet. Key words: Plasmodiumfalciparum, Sulfadoxine, Resistance, A437G dhps gene
