DNA BARCODE IKAN SENGGIRINGAN (Desmopuntius sp.) BERDASARKAN GEN SITOKROM C OKSIDASE SUBUNIT I (COI)
Striped barb (Desmopuntius sp.) is one of the freshwater fishery resources that are widespread in Sumatra. Overfishing, the use of non-selective fishing gear, and the presence of water pollution are feared to result in a reduction of striped barb population. The purposes of this study were to obtain the mitochondrial DNA cytochrome C oxidase subunit I (COI) gene sequence, determine the percentage of identity, genetic distance and the phylogenetic between species of striped barb and to determine the physics and chemical properties of the aquatic habitat at Ogan and Lematang Rivers in the South Sumatra. This research will be carried out in October 2021-Januari 2022. The method used in barcoding are isolation of DNA, electrophoresis, DNA amplification using PCR (Polymerase Chain Reaction), sequencing of COI gene and phylogenetic trees construction using the Neighbor Joining (NJ) method as well as measurement of water quality of striped barb habitat. The sequence of COI mtDNA gene fragments were obtained from PCR results with an annealing temperature optimization of 50.4°C for 30 seconds in 39 cycles. The nucleotides length of the COI gene of striped barb was 681 base pairs (bp). The results of BLASTn analysis of striped barb samples from Lematang River and Ogan River had the highest similarity with Desmopuntius gemellus from Sumatra and Kalimantan of 99.33% (Lematang River) and 99.50% (Ogan River). Phylogenetic of striped barb from Lematang River and Ogan River is in the same cluster as Desmopuntius gemellus.
