Clownloach fish (Chromobotia macracanthus) is a type of ornamental fish belonging to the Cobitidae tribe. Botia fish in Indonesia live in Sumatra (the Batanghari Jambi and the Musi-Ogan rivers of South Sumatra). Genetic studies of botia fish species from Sumatra are out of exploration. DNA (Deoxyribonucleic acid) barcoding programs are needed as genetic conservation, cultivation and domestication purposes that can preserve botia fish in nature. This study aims to determine the nucleotide sequence of the COI (Cytochrome C oxidase subunit I) gene. These research methods include DNA isolation, DNA amplification using PCR (Polymerase Chain Reaction), sequencing, constructed phylogenetic and genetic distance and water quality measurement. The results of this study indicated that samples of 10 juvenile botia fish have morphometric measurements of total length average from 13-14 cm, a standard length of 8,9 cm and meristic. measurements were, 11 dorsal (D) fingers, 17 caudal ( C), 8 anal (A), 16 pectoral (P), 9 ventral (V), 8 antennae. The size of the DNA band was 636 bp with annealing temperature 51°C. The results of BLASTn LK 2,3,4,5. dan SK 1,2,3,5 had the highest similarity with Indian C. macracanthus (98.80%), except on LK 3 (98,11%). The genetic distance 0.000 and the phylogenetics tree had a BV (Bootstrap value ) 98% againts the same genus of C. macracanthus. The results of the measurement water quality parameters from 2 rivers ie, temperature ranged from 29.8-31.1° C, pH 5.50-7.20, DO 5.47-6.76 mg L-1, ammonia 0.0189-0.2115 mg L-1, TDS 0.012- 0.172 mg L-1, salinity 0.5 ppt, total alkalinity 8-18 ppm, water transparrent 20-38.5 cm, and current velocity 2.9-10.2 cm s-1.
