ANALISIS IN VITRO: PENENTUAN KONSENTRASI EKSTRAK DAUN TERATAI (Nymphaea pubescens L.) SEBAGAI KANDIDAT OBAT MOTILE AEROMONAS SEPTICEMIA (MAS)
One of the freshwater bacteria that causes economic losses in freshwater fish farming is Aeromonas hydrophila because it can cause Motile Aeromonas Septicemia (MAS) disease. The use of lotus leaf extract as an antibacterial agent in Aeromonas hydrophila resulted in cell wall lysis and atrophy. This study purpose to determine the concentration of lotus leaf extract (Nymphaea pubescens L.) to inhibit and kill Aeromonas hydrophila bacteria in vitro. This research was conducted at the Aquaculture Laboratory and Experimental Pond, Aquaculture Study Program, Faculty of Agriculture, Sriwijaya University in January – March 2022. This study used a post only control group design. Which compared the antibacterial effect of lotus leaf extract with control and treatment with 3 repetitions. Parameters observed include determining the value of Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC) and analysis of phytochemical tests. The results of this study showed that lotus leaf contains alkaloids, flavonoids, tannins and saponins. The MIC value of lotus leaf extract showed that it was included in the very strong category, namely at a concentration of 600 mg L-1 with an inhibition zone value of 22.61 ± 0.15 mm. The value of MBC is known to be good because there were no bacterial colonies growing on the medium with a concentration of 491,60 mg L-1.
