Background: Sepsis is a complex clinical syndrome that results from a harmful or damaging host response to infection. Sepsis ranks at the top 10 leading causes of death in United State. Incidence of sepsis increases every year. Sepsis releases inflammatory mediators and actives of the excess. There are a variety of cytokines that play a role in the inflammatoiy process, the most important, TNF-a, IL-1, IL-6, IL-8, IL-12 as a proinflammatory cytokine and IL-10 as antiinflammatory. Interleukin 10 plays a role in limiting and terminating the inflammatory response. Excessive IL-10 can cause immune system suppression and increased mortality. Interleukin 10 gene pplymorphisms are known risks and outcomes associated with sepsis. Mutations in genes that encode interleukin 10 gene influence interleukin 10 gene transcription and interleukin-10 production rpsulting in increased levels of interleukin 10, Elevated levels of interleukin-10 is the cause that will lead to increased risk of sepsis and fatal sepsis output. This study aims to identify the promoter polymorphism -1082 A/G interleukin-10 gene in patients with sepsis in surgical wards and ICUs RSUP. Mohammad Hoesin Palembang. Methods: This was a descriptive observational study with cross sectional sample of 48 patients with sepsis. Identification of -1082A/G polymorphism interleukin 10 gene was done by using ARMSPCR (AmplificcUion Refractory Mutation System). Results: The frequency of aged over 40 years patients as much as 54J 7% and aged under 40 as much as 45,83%. The average age of patients is 41.02 years. The frequency of male were 62,5% and female 37,5%. The source of infections come from digestive tract as much as 45,8% and urogenital tract 22,9% , skin and soft tissue 12.5%, orthopedic 12.5% and thoracic 6.2%. Most samples came from the tribe of Palembang as much as 31,2%, Sekayu 18,7%, Komering 14.5%, Muara Enim 8.3%, Semendo (Lahat) 6.2%, Ogan 6.2%, Lematang (Pagar Alam) 6.2%, Java 4.2%, Prabumulih and Lubuk Linggau 2.1%. The genotype AA was found as much as 58,3%, 37,5% AG genotype, and 4,2% genotype GG. Frequency of A alel is 77,08% and G alel is 22,92%. Conclusion: The overview of wild-type genotype were more common in patients with sepsis who was treated at surgical ward and ICU RSUP. Mohammad Hoesin Palembang period January to May 2011 (58,3%).
