SKRINING BAKTERI PENGHASIL SELULASE ASAL TANDAN KOSONG KELAPA SAWIT
Empty fruit bunch of palm oil contains cellulose which can be used as a substrate for the growth of cellulolytic bacteria to produce cellulase enzyme. This research aimed to determine the bacteria that have potential to degrade cellulose from empty fruit bunch of palm oil and to determine the characteristics of bacteria to cellulase enzyme producing. This research is an exploratory descriptive study, the results are presented in tabular form. Isolation of cellulolytic bacteria was done using selective media Carboxy Methyl Cellulose (CMC 1%) and then screening using 0,1% Congo red test the cellulolytic potential which was marked by the appearance of a clear zone around the colony. The five selected isolates had the largest cellulolytic activity index was in isolates 41, 37, 6, 42, 30 namely 1.95 mm, 1.65 mm, 1.5 mm, 1.18 mm and 1.05 mm. Quantitative test using DNS spectrophotometry method showed that the highest enzyme activity was in isolates 41, 37, 6, 42, 30 namely 0.392 U/mL, 0.366 U/mL, 0.356 U/mL, 0.360 U/mL and 0.347. U/mL. Five isolates had a characteristic round colony and white colony shape and five isolates are Gram-positive bacteria with bacil cell form.
