The development of advances in reproductive technology in livestock, one of which is spermatozoa sexing which can separate X and Y chromosome spermatozoa which aims to increase reproductive efficiency in running a cattle business. The decrease in the quality of spermatozoa after sexing is common due to the centrifugation process. This study was conducted with the aim of evaluating the 5-10% albumin column sexing method with different incubation times and no centrifugation process was carried out to improve the quality of spermatozoa. This research was conducted from August to October 2021 at the Forage Breeding Center (BP-HPT) Sembawa Banyuasin, South Sumatra. The material used in this study was fresh semen of simmental cattle which was treated with incubation time, namely P1 = incubation time of 40 minutes, P2 = incubation time of 50 minutes, P3 = incubation time of 60 minutes and carried out 4 times repetitions. The design used in this study was a Completely Randomized Design (CRD) method and a further test of variance table was carried out, where the parameters observed were the quality of fresh semen, motility and viability before freezing (BF) and after diluting/Post Thawing (PT). ). The results showed that the test according to the motility and viability variance table in each treatment BF and PT was not significantly different (P> 0.05). There was a drastic decrease from BF to PT after freezing the semen resulting from sexing, for the highest motility and viability values were achieved by P1 BF with a motility value of X spermatozoa of 60% and a viability value of 65.3% while Y spermatozoa had a motility value of 61, 25%, and the viability value is 63.84%. So that the environmental temperature required for spermatozoa greatly affects the motility and viability of spermatozoa where the best time for incubation required for sexing spermatozoa using the 5-10% BSA method is 40 minutes.
