ANALISIS KADAR NIKOTIN PADA TEMBAKAU (NICOTIANA TABACUM L) MENGGUNAKAN METODE GC-MS
Nicotine is an alkaloid compound found in tobacco. Nicotine content is included in the tobacco quality parameters. The nicotine content is 1-3% based on the quality requirements according to SNI 0612-1989-A. This study aims to determine nicotine levels in mol and Ranau tabacum varieties using gas chromatography-mass spectrometry (GCMS). The study begins with a phytochemical study of alkaloids using the Dragendorff's reagent. In addition, the alkaloid test was performed by thin layer chromatography (TLC) using ethyl acetate: methanol (6: 4; 7: 3; 8: 2) in the stationary and mobile phases (eluents) of silica gel. bottom. Under GCMS operating conditions, a TG5MS column (30m x 0.25mm) 0.25µm thick was used and helium was used as the mobile phase at a flow rate of 1mL / min. The results of the phytochemical screening showed a positive test for alkaloids in the presence of a red brick-colored precipitate. TLC tests also showed the presence of alkaloid compounds with red brick stains. GCMS results showed the presence of several alkaloid compounds such as nicotine, nornicotine and anatabine. The results of the tobacco extract test showed that the molar and Ranau varieties had nicotine levels of 0.02 g and 0.06 g respectively. The nicotine content produced in tobacco samples is very low and is not included in the quality requirements of SNI 0612-1989-A
