UJI AKTIVITAS ENZIM AZOREDUKTASE EKSTRASELULER DARI BAKTERI PSEUDOMONAS STUTZERI
Extracellular azoreductase enzyme activity test has been carried out from the Pseudomonas stutzeri. P. stutzeri is bacteria bacteria that have the ability to decolorize azo dyes against methyl red dyes.This study aimed to examine the activity of the extracellular azoreductase enzyme from P. stutzeri. The azoreductase enzyme extract from P. Stutzeri was obtained by centrifugation of the liquid culture on NB media which was incubated for 2x24 hours at 37◦C in an incubator shaker. The results of the selected fractions were used to calculate enzyme activity and protein content. Determination of the optimum conditions for the extracellular azoreductase enzyme was carried out at variations in incubation time, pH, temperature, substrate concentration and calculation of enzyme kinetics Vmax and Km. The highest fraction was fraction 3 with a saturation level of 40%-60% with an enzyme activity of 0.1101 Unit/mL and a specific enzyme activity of 0.1079 U/mg. Extracellular Azoreductase enzyme from P. stutzeri has optimum conditions at 5 minutes incubation time, pH 7, temperature 37ºC and substrate concentration 70µM with enzyme activity 0.1166 U/mL. The values of Vmax and Km obtained are 0.173683 U/mL and 31,33415 M, respectively.
