ISOLASI YEAST DARI AIR KELAPA YANG DIGUNAKAN SEBAGAI AGEN MIKROBA PADA FERMENTASI SUBSTRAT GLUKOSA, XILOSA DAN ARABINOSA MENGHASILKAN ETANOL
Research on the isolation and identification of coconut water yeast used as a source of microbial agents in assisting fermentation to produce ethanol has been done. The isolates selected were coconut water isolates from a dilution of 10-4 which were spread in YMA media which produced 48 yeast isolate colonies which were then selected based on macroscopic physiological differences to produce 20 yeast isolates with different textures, colors and shapes, where the selected isolates these were named AK 1 to 20. Through the phenotypic test using medium with different substrates (glucose, xylose and arabinose) at 30oC and 42oC, 5 yeast isolate colonies were found that could grow well on these media. The yeast isolate that have grown were further analyzed by genetics with the VITEX® 2, an automated compact system. The isolates were as identifiedwhichCandida tropicalis, Candida lusitaniae, Cryptococcus laurentiiand Candida famata.Isolates used for the fermentation of glucose, arabinose and xylose substrates. The highes ethanol concent produced by fermentation of glucose byAK 18 (Candida famata) was 2.5310%, on xylose and arabinose substrates, the highest ethanol content was found in isolate AK 12 (Cryptococcus laurentii) with ethanol content of 1.5233% and 1.3682%, respectively.
