Molecular markers are the key to success in DNA barcoding techniques. One of the use of molecular markers which until now has not been widely used in research is trnK. The use of specific genes in molecular techniques will produce accurate data in identifying and knowing the genetic information of a species. The genetic information of red rice, which is currently threatened with extinction, especially the local varieties of South Sumatra, is very limited and even relatively lacking. The research methods carried out were DNA isolation, electrophoresis of DNA isolation results, DNA amplification using PCR, DNA sequencing, data analysis and data presentation. This study used 8 samples of brown rice from different places of life, namely Keli Rejo brown rice, Cahya tani, Bungin Tinggi, Linggau, Jaya Mulya, Sirah Pulau Padang, Suak Batok and Sumber jaya. The primer used was trnK primer with annealing temperature of 58.30C. The results showed that the quantitative test of DNA samples from Bungin Tinggi, Linggau, Jaya Mulya, Suak Batok and Sumber Jaya with OD values ranging from 1.89 to 1.91, and the results of isolated DNA samples with the largest concentration, namely Sirah Pulau Padang sample, which was 44 ,9 ng/μl, and the smallest concentration is Bungin Tinggi with a value of 6.7 ng/μl. The result of the blast process with the NCBI website is that there are no similarities or the existence of the genbank database is not found. The conclusion of this study is that the trnK gene analysis has not been successful because of several obstacles that occurred in the early stages, namely DNA isolation in the form of impure DNA isolation results and many contaminants visualized through thin DNA bands and smears. These factors can be influenced by the condition of the leaf samples that are not good, the DNA isolation process is not perfect and the PCR profile that needs to be done, for example the anneling temperature, adding the reaction cycle and the time in the DNA amplification process with PCR tools.
