EFEKTIVITAS METODE GEL CLOT DAN CHROMOGENIC PADA UJI BAKTERI ENDOTOKSIN MENGGUNAKAN DARAH HORSESHOE CRAB ASAL BANYUASIN, SUMATRA SELATAN
Mimi or horseshoe crab is one of the marine animals that is included as a rare animal with an endangered status and is found in the waters of Banyuasin Estuary, South Sumatra. The blood of horseshoe crab has the ability to detect bacteria, which in the blood contains amebocyte lysate which plays an important role in the detection of bacterial endotoxins, so research is carried out on the ability of horseshoe crab blood from Banyuasin Estuary waters, South Sumatra in detecting endotoxin bacteria using the Gel Clot method and Chromogenic, and comparing their effectiveness. Sampling was carried out in Banyuasin Estuary waters, then sample testing was carried out at the Marine Bioecology Laboratory, Faculty of Mathematics and Natural Sciences, Sriwijaya University and the Laboratory of Applied Genetic Engineering and Protein Design, Indonesian Institute of Sciences Biotechnology Research Center, Bogor. The results of the endotoxin test showed that male and female C. rotundicauda, and male T. gigas were able to detect bacterial endotoxins using the Gel Clot method and the Chromogenic method. Gel Clot method is more effective in detecting endotoxin bacteria by successfully detecting 4 (four) blood samples from 28 blood samples, with a method that is easier to perform. While the Chromogenic method can only detect 1 (one) sample from 28 samples with a correlation value r = 0.905 using a more difficult method.
