IDENTIFIKASI MUTASI GEN DHFR Plasmodium falciparum KODON S108N SEBAGAI MARKER RESISTENSI MALARIA TERHADAP PIRIMETAMIN DI SUMATERA SELATAN
Background:Malaria was still a cause of morbidity and mortality in infants, children and adults (especially pregnant women), especially in tropical countries such as Indonesia. This study aims to identified the mutation in the DHFR gene of Plasmodium falciparum to pyrimethamine were cut with enzymes Bsrl Plamodium falciparum malaria in patients in the dr. Mohammad Hoesin Palembang hospital, Sekayu hospital, Lahat hospitals, Baturaja hospital and clinic laboratories in South Sumatra. Methods:This study was a descriptive study of 30 samples of patients with Plasmodium falciparum malaria. Identification of Plasmodium falciparum DHFR gene mutations was done by using Polymerase Chain Reaction (PCR) amplification and Restriction Fragment Length Polymorphism (RFLP) using enzyme Bsrl. Results:ln patients with Plasmodium falciparum malaria in dr. Mohammad Hoesin Palembang hospital, Sekayu hospital, Lahat hospital and Baturaja hospital, and clinical laboratories who being the subjects of studies, the result were 2 people (33.3%) were homozygot mutated DHFR gene of Plasmodium falciparum to pyrimethamine, whereas 4 others (66.6%) without the mutation (wild-type) of the 30 isolates subjects of study. ConclusionrGenotype of mutant 2 (33.3%), genotype wild-type (normal) 4 66.6% of the 6 positive samples amplified from 30 isolates. Pyrimethamine was still adequate for the treatment of Plasmodium falciparum malaria in South Sumatra.
