Drying method is a way to decrease the moisture content of material. The drying on the plant greatly affects the bioactive compound produced. This research aimed to determine the effect of different drying methods on the yield,total polyphenols content, total flavonoids content and antioxidant activity on the water lettuce (Pistia stratiotes) leaves extract. This research was carried out experimentally in a laboratory with a drying method consisting of 3 treatments (sun drying, oven drying, and freeze drying) and repeated 3 times. Parameters were measured in this study including extract yield, bioactive compounds (polyphenols and flavonoid) and antioxidant activity. The values obtained were described in the form of pictures, graphs, and continued with DMRT test on 5% level. The results showed that the yield of the water lettuce leaves extract with the drying method sun-drying, oven-drying and freeze-drying respectively was 8,48%, 13,54% and 15,64%. The total content of polyphenols in the water lettuce leaves extract with the drying method sun-drying, oven-drying and freeze-drying respectively was 12,95 mg GAE/g sample dry sample, 18,33 mg GAE/g dry sample and 32,93 mg GAE/g dry sample. The total content of flavonoids in the water lettuce leaves extract with the drying method sun-drying, oven-drying and freeze-drying respectively was 58,80 mg QE/g dry sample, 99,57 mg QE/g dry sample, and 146,80 mg QE/g dry sample. Antioxidant activity using the DPPH method resulted in the IC50 value of the water lettuce leaves extract with the drying method sun-drying, oven-drying and freeze-drying respectively was 462,67 ppm, 305,67 ppm and 266,33 ppm. The method that produces the best value for the total content of polyphenols, flavonoids and antioxidant activity of apu-apu leaves extract is the oven drying method.
