Antioxidants are chemical compounds that can protect body cells from damage caused by free radicals. Antioxidants based on the source are divided into two, namely natural antioxidants derived from plants and synthetic antioxidants made from chemicals. Natural antioxidants are obtained from plants such as mangrove leaves and become a potential alternative to be developed as a substitute for synthetic antioxidants. The Musi River Estuary area is dominated by mangrove forests, one of which is the Avicennia alba mangrove because it is located at the confluence of fresh water and sea water so that the salinity around it is quite high and is classified as an estuarine ecosystem (brackish waters). The purpose of this study was to determine and analyze the potential activity of antioxidant compounds qualitatively and quantitatively crude extract and after isolation from A. alba mangrove leaves. The antioxidant activity test method uses DPPH reduction, while the isolation method used is column chromatography and thin layer chromatography. The results of antioxidant activity from several qualitative test concentrations on crude extracts and isolates changed color from purple to yellow so that they had antioxidant potential. Quantitative analysis of crude extract has a high potential in methanol extract based on the IC50 value of 78 ppm. After being isolated, the methanol extract showed a very strong antioxidant potential in fraction 9 which was eluted with 5:5 eluent and the isolate was brownish green with an IC50 value of 40 ppm.
