Isolation and ability test of non symbiotic nitrogen fixing bacteria from mangrove area of Sembilang National Park, South Sumatera had been done from March to May 2014. Samples of soil taken from the mangrove forests of Sembilang National Park, South Sumatera by cluster sampling method. Isolation, selection, characterization, and identification were done in the Laboratory of Microbiology, Department of Biology, Faculty of Mathematics and Natural Science. Analysis of total Nitrogen, was done in the Laboratory of Plant Physiology, Faculty of Agriculture, Sriwijaya University. The objective of the research was to obtain non symbiotic nitrogen fixing bacteria isolates from mangrove rhizosphere soil, obtained the name of the genus with an index of similarity based on the characteristics and determine its ability to tie up nitrogen. It was obtained 21 isolates of bacteria from selection results which were able to form pellicle on the surface of the N-free Na malate media. The results of ability test was obtained 6 bacterial isolates that have the largest colony diameter of three stations. Based on dendogram, ST1P1K2 showed the similarity index isolate of 78% with genus of Planococcus, ST1P1K4 have 50% similarity with genus of Erwinia, ST1P2K1 have 69% similarity with genus of Azotobacter, ST1P5K1 have 94% similarity with genus of Azotobacter, STiP6K4 have 38% similarity with genus of Brucella and ST2PiK2 have 76% similarity with genus of Derxia.
