Isolation, characterization, and identification of non-symbiotic nitrogen-fixing bacteria was performed to obtain the isolates of non-symbiotic nitrogen-fixing bacteria from ultisols soil and to identify the genus. This research was conducted ffom February until August 2014 in the laboratory of Microbiology, Biology major, faculty of Mathematics and Natural Science, Sriwijaya University. Sample of ultisols soil were taken from the regions of Ogan Ilir, Prabumulih and Kayu Agung. Soil used as sample to analyzed were taken at depths of 0, 15 and 30 cm. Isolation were done with the use of medium Nutrient Agar (NA) and the selection was done with the use of medium LGI. After that, the characterization process began. This included the colony morphology, bacterial cells, and the nature of the bacterial physiology. The study design used was Completely Randomized Design (CRD) with the treatment using types of isolates. Data analyzed were using analysis of variance in Statistics program version 8.0. If the result showed significant difference, then Duncan New Multiple Range Test (DNMRT) conducted with a 0.05. The basic characters of isolates then matched with the chosen genus. After that, dendogram created by using cluster analysis to determine the percentage of similarity with the chosen genus. From the analysis, it was found that there were 8 bacterial isolates that have high potential to tie up free nitrogen in the air within the biggest colony diameter of 9.72 mm by isolate P9. Based on the result of cluster analysis that isolate Ki had the similarity percentage with genus Dencia 82%, isolate K3 with ganus Beijerinckia 54%, isolate O3 with genus Erwinia 46%, isolate 06 with genus Pseudomonas 83%, isolate P2 with genus Dencia 82%, isolate P3 with genus Bacillus 64%, isolate P8 with genus Dencia 48%, and isolate P9 with genus Derxia 89%.
