Motile Aeromonas Septicemia (MAS) is common diseases occoured in freshwater fish. The disease is caused by the bacterium Aeromonas hydrophila. The mortality rate ranges caused by MAS infection in fish from 80-100% within 1-2 weeks (Kusen et al., 2015). Treatment of A. hydrophila infection be done by giving antibiotic, but it causes residues in the environment, fish and results in resistance to bacteria (Le et al., 2018; MKPRI, 2014). The use of garlic (Allium sativum) and bilimbi (Averrhoa bilimbi) in the treatment of fish disease has been shown to inhibit the growth and kill bacteria that infect fish because the plant has antibacterial properties and effectively stimulates the fish immune system (Erguig et al., 2015; Purnamasari et al., 2015; Dwinanti et al., 2019; Zhou et al., 2016). The researchers aimed to examine the combination effects of garlic and bilimbi as drug candidates for MAS disease by in vitro analysis. This research is expected to be able to produce effective drug preparations to treat MAS disease in fish. This research was conducted in Aquaculture Laboratory, Aquaculture Study Program, Microbiology Laboratory and Biotechnology, Fishery Products, Faculty of Agriculture Sriwijaya University from October 2020 until January 2021. The research tested comparing the combination of garlic (G) and bilimbi (B) at different concentration of 5 treatments (T) and 3 repeatations. The treatments are 100% garlic (T1), combination of 75% G and 25% B (T2), combination of 50% G and 50% B (T3), combination of 25% G and 75% B (T4), bilimbi B 100% (T5). Parameters was phytochemical contents (tannin, saponin, flavonoid and alkaloid), Minimum Inhibitory Concentration (MIC) value, Minimum Bactericidal Concentration (MBC) and Effective Concentration 50% (EC50) values. The results showed that each treatment has active compounds of tannin, saponin, flavonoid, and alkaloid. MBC values are only obtained at T1 and T2 at 100% concentration. MIC treatment values for each treatment is T1 4%, T2 6%, T3 12%, T4 12% and T5 36%. While the EC50 values of each treatment is T1 6.2%, T2 36.8%, T3 26.6%, T4 30.9% and T5 32%. The conclusion of this study is that the combination of garlic and bilimbi does not eliminate the active ingredients present in a single solution of garlic and bilimbi. Tannin, saponin, flavonoid and alkaloid remain detected in either combination or single solutions in all treatments. The best antibacterial activity in combination treatment is the P2 treatment (garlic 75% + bilimbi 25%) which is indicated by a lower Minimum Inhibitory Concentration (MIC) value than other combinations, which was shown to be smaller than other combinations, as well as the Minimum Bactericidal Concentration (MBC) value obtained only in the P2 treatment. The EC50 value obtained is 36.8% for MAS disease can be continued to the in vivo test.
