Actinomycetes are a group of Gram positive bacteria and have been widely recognized as microorganisms that produce many bioactive compounds. Actinomycetes are widely distributed in nature, especially in the soil, one of which is swampland. Swamp soil usually has a low soil pH and has an abundant source of organic matter so it is good for supporting microbial life that decomposes organic matter, one of which is the actinomycetes group. The bioactive compounds produced by actinomycetes have many benefits that can be used as antifungal, antibacterial, anti-tumor, and others so that there is a high chance for researchers to conduct further research on actinmycetes, especially the genus Streptomyces which has been widely known as a producer of antibacterials. The research was conducted from December 2020 to April 2021, in the Microbiology laboratory and Genetics and Biotechnology laboratory, Deparment of Biology, Faculty of Mathematics and Natural Science, Sriwijaya University. This study was conducted to determine the actinomycetes isolated from marshland have the potential as an antibacterial against Escherichia coli ATCC25922 and Staphylococcus aureus ATCC25923, macroscopic, miscroscopic, and biochemical characterization, and identification of actinomycetes. The conclusion from this research is that 19 isolates of actinomycetes obtained from swamp soil have the potential to produce antibacterial because they can inhibit the growth of Escherichia coli ATCC25922 and Staphylococcus aureus ATCC25923. One of the actinomycetes isolate (ACTI 8) had antibacterial activity against Escherichia coli ATCC25922 and Staphylococcus aureus ATCC25923. Meanwhile, 3 other actinomycetes isolates (ACTI 10, ACTI 11, ACTI 13) had antibacterial activity only on Staphylococcus aureus ATCC25923. After characterization, these four potential actinomycetes isolates belonged to the genus Streptomyces. Keywords: Actinomycetes, Antibacterial, Streptomyces, Swampland. Actinomycetes adalah kelompok bakteri Gram positif dan telah banyak dikenal sebagai mikroorganisme yang banyak menghasilkan berbagai senyawa bioaktif. Actinomycetes terdistribusi luas di alam terutama di tanah, salah satunya tanah rawa. Tanah rawa biasanya memiliki pH tanah yang rendah serta memiliki sumber bahan organik yang melimpah sehingga baik untuk mendukung kehidupan mikroba pengurai bahan organik salah satunya kelompok actinomycetes. Senyawa bioaktif yang dihasilkan actinomycetes memiliki banyak manfaat yang dapat digunakan sebagai antijamur, antibakteri, anti tumor dan lainnya sehingga berpeluang tinggi bagi peneliti untuk melakukan penelitian lebih lanjut terhadap actinomycetes, terutama Genus Streptomyces yang telah banyak dikenal sebagai penghasil antibakteri. Penelitian ini telah dilakukan dari bulan Desember 2020 sampai dengan April 2021, bertempat di Laboratorium Mikrobiologi dan Laboratorium Genetika dan Bioteknologi, Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Sriwijaya. Penelitian ini dilakukan untuk mengetahui actinomycetes yang diisolasi dari tanah rawa berpotensi sebagai antibakteri terhadap Escherichia coli ATCC25922 dan Staphylococcus aureus ATCC25923. Tahapan penelitian yang dilakukan yaitu isolasi, pemurnian, uji antibakteri terhadap Escherichia coli ATCC25922 dan Staphylococcus aureus ATCC25923, karakterisasi secara makroskopis, mikroskopis dan biokimia serta identifikasi actinomycetes. Kesimpulan yang didapatkan dari penelitian ini adalah actinomycetes yang didapatkan dari tanah rawa berjumlah 19 isolat dan diantaranya berpotensi menghasilkan antibakteri karena dapat menghambat pertumbuhan dari Escherichia coli ATCC25922 atau Staphylococcus aureus ATCC25923. Salah satu isolat actinomycetes (ACTI 8) memiliki aktivitas antibakteri terhadap bakteri Escherichia coli ATCC25922 dan Staphylococcus aureus ATCC25923. Sedangkan 3 isolat actinomycetes lainnya (ACTI 10, ACTI 11, ACTI 13) memiliki aktivitas antibakteri hanya pada bakteri Staphylococcus aureus ATCC25923. Setelah dilakukan karakterisasi keempat isolat actinomycetes yang berpotensi ini termasuk dalam genus Streptomyces. Kata Kunci : Actinomycetes, Antibakteri, Streptomyces, Tanah Rawa.
