PENGARUH SUMBER KARBON DAN NITROGEN PADA BIODEGRADASI ZAT WARNA DIRECT RED 80 MENGGUNAKAN Aeromonas jandaei BD 02 dan Pseudomonas guguanensis BD 14
Kain Jumputan Palembang is a commodity that is growing rapidly along with the large number of consumer demands from various parts of Indonesia and abroad. About 60-75% of synthetic dyes used in the textile industry are azo dyes. Azo dyes such as Direct Red have a complex structure, are soluble in water, contain metals, have an aromatic structure and have one or more azo bonds (–N=N–) making them difficult to degrade. The biological method is an effective dye removal method because it does its job quickly, is cost-effective, environmentally friendly and does not produce secondary contaminants. The presence of azoreductase enzyme is thought to play a role in breaking azo bonds. The need for carbon and nitrogen in the biodecolorization process is very necessary to increase the ability of the bacteria Aeromonas jandaei BD 02 and Pseudomonas guguanensis BD 14 in degrading Direct Red 80 dye. Microorganisms need different carbon and nitrogen sources to increase the ability of biodegradation, growth and survival of microorganisms so that able to produce azoreductase enzymes that play a role in breaking azo bonds in Direct Red 80 dye. The research was carried out from February 2021 to May 2021, at the Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. This study used a factorial Randomized Block Design with 3 treatment factors. Factor I is the type of bacteria (Aeromonas jandaei BD 02 and Pseudomonas guguanensis BD 14), factor II is the type of carbon source (glucose, sucrose, and lactose) and factor III is the type of nitrogen source (yeast extract, peptone and tryptophan). The experiment was repeated three times test. Based on the research that has been done, it can be seen that, the addition of a 1% lactose carbon source with a nitrogen yeast extract of 0,5% resulted in the best number of bacterial cells in Aeromonas jandaei BD 02 bacteria reaching 4,6 x 1010 cfu/mL and Pseudomonas guguanensis BD 14 reaching 5,2 x 109 cfu/mL. The addition of a carbon source 1% sucrose with tryptophan 0,5% as nitrogen source produced the highest percentage of degradation power in Aeromonas jandaei BD 02 of 86,66% and Pseudomonas guguanensis BD 14 of 84,60%. The results of biodegradation of Direct Red 80 dye produced different chromatogram patterns. The Rf value in the control was 0,89 and in the treatment culture Aeromonas jandaei BD 02 + Sucrose + Tryptophan had an Rf value of 0,62 while Pseudomonas guguanensi BD 14 + Sucrose + Tryptophan was 0,56. The addition of carbon and nitrogen sources had a positive effect on the growth of Aeromonas jandaei BD 02 and Pseudomonas guguanensis BD 14 bacteria and increased the percentage of Direct Red 80 dye degradation.
