Musi River is a river that becomes the estuary of dozens of rivers in both Bengkulu and South Sumatra, has a length of about 750 km across the city of Palembang. The estuary of the Musi River is one of the places used by the community as a shipping lane, fishing area for fishing resources, waste disposal and other human activities. The amount of input of organic matter either directly or indirectly will affect the condition of the waters, including the effect on the abundance of plankton. One of the biotic components that determine life in the waters is plankton. Plankton includes two major groups, namely phytoplankton which are plant and zooplankton which are animal. The purpose of this study was to analyze the diversity and abundance of plankton in the Musi River Estuary, South Sumatra. The research method used is a purposive sampling technique, which is a sampling technique based on the representation of the location of the waters. The plankton samples obtained were preserved with 4% formalin and then identified using a reference book, then the diversity was calculated and the abundance analyzed. The results of the calculation of the Diversity Index (H') of plankton during the study ranged from 1,310-1846, with 17 genera found, 7 classes consisting of 253 individuals. It can be concluded that the diversity of plankton at the mouth of the Musi River is classified as moderate. Based on the composition, the phytoplankton of the Bacillariophyceae class dominates with a percentage of more than 50%, this occurs because the phytoplankton of this class are able to adapt to the environment in which they live. The abundance of plankton in the Musi River Estuary ranges from 7-29 ind/l where the highest abundance is at station 1. Physical parameters measured were brightness values ranging from 26-40 cm, current velocity ranging from 0.91 to 1.11 m/s including medium speed and water temperature analysis ranging from 29.2 to 30.9 °C. The chemical parameters are pH with a value of 4.2-7, DO ranging from 5.33-6.5 mg/l, phosphate with a value of 0.62-2.04 mg/l, nitrate 0.8 mg/l-2, 26 mg/l, while the salinity value is 0. Measurement of the physical and chemical parameters of the waters around the Musi River Estuary is still considered optimal and good for supporting the survival of plankton.
