Sembilang fish (Plotosus canius, Hamilton: 1822) is one of the species of fish consumed by the community around the Musi River Estuary, Sungsang Village, Kec. Banyuasin II, Kab. Banyuasin. The increasing need for Sembilang fish consumption can decrease the fish population in that area, so it is necessary to research to obtain information about the Reproductive Biological Aspects of Sembilang Fish around the Musi River Estuary, Sungsang Village, Kec. Banyuasin II, Kab. Banyuasin. This research aims to analyze the relation of length and weight fecundity, sex ratio, gonadal maturity level (TKG), Gonad Maturity Index (IKG), fecundity, egg diameter, and spawning patterns of the Sembilang fish. This research was conducted from February-May 2021 at the Biosystematics Laboratory of the Department of Biology, Faculty of Mathematics and Natural Sciences. The research procedure begins with cencused by fisherman. This is a sampling technique that provides equal opportunities for each fish to take each element of the fish population. Sembilang fish from fishermen's catches obtained 30 fish, and only 12 fish were gonadally mature. Then samples were censused for all parts of the gonads for analysis. The samples measured included the total length of fish, body weight, gonad weight, and egg diameter. The parameters observed included the level of gonad maturity and sex ratio. Furthermore, all fish samples were censused for analysis. The relation of length and weight fecundity was analyzed using the regression method and the gonadal maturity index was analyzed using the quantitative method.The results showed that the correlation value (r) of the length-fecundity relationship was 0.62 and the weight-fecundity relationship was 0.72. The Sex Ratio Comparison was 1:2.3. The gonadal maturity level (TKG) obtained during the research was the most common in Phase I TKG with a range of 0.04-17.9% Gonad Maturity Index (GI), and the Fecundity of Sembilang Fish can reach 690-2514 eggs. The size of the fish egg diameter varies with the range of 3.01-4.12 mm and the spawning pattern grouped in stages. So it can be concluded that the Sembilang fish obtained tend to be in TKG I with a Sex Ratio of 1:2.3 and are unbalanced group. The resulting fecundity ranged from 690-2514 with partial or continuous spawning patterns. Suggestions from research on reproductive biology aspects of fish are should be done every month to determine TKG, fecundity, and egg diameter size, so that the spawning time can be known and it can complement the existing information. Ikan Sembilang (Plotosus canius, Hamilton:1822) merupakan salah satu jenis ikan yang dikonsumsi masyarakat di Sekitar Muara Sungai Musi Desa Sungsang Kec. Banyuasin II, Kab. Banyuasin. Meningkatnya kebutuhan konsumsi ikan Sembilang berpotensi untuk menurunnya populasi ikan ditempat tersebut sehingga perlu dilakukan penelitian untuk mendapatkan informasi mengenai Aspek Biologi Reproduksi Ikan Sembilang di Sekitar Muara Sungai Musi Desa Sungsang Kec. Banyuasin II, Kab. Banyuasin. Penelitian telah dilaksanakan pada Bulan Februari-Mei 2021 bertempat di Laboratorium Biosistematika Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam. Pengambilan sampel dilakukan oleh nelayan dengan metode sensus yaitu teknik pengambilan sampel yang memberikan kesempatan yang sama pada setiap individu ikan sembilang untuk diambil setiap elemen populasi ikan tersebut. Ikan Sembilang dari tangkapan nelayan didapatkan 30 ekor dan hanya 12 ekor yang sedang matang gonad kemudian sampel di sensus semua bagian gonadnya untuk dianalisis. Sampel yang diukur meliputi panjang total ikan, bobot tubuh, bobot gonad dan diameter telur, parameter yang diamati meliputi tingkat kematangan gonad, dan rasio kelamin. dan sampel ikan disensus semua untuk di analisis. Hubungan Panjang berat –fekunditas di analisis menggunakan metode regresi dan Indeks kematangan gonad dianalisis menggunakan metode kuantitaif. Hasil dari penelitian didapatkan bahwa Nilai korelasi (r) Hubungan panjang-fekunditas sebesar 0,62 dan hubungan berat-fekunditas sebesar 0,72, Perbandingan Rasio Kelamin yaitu 1:2,3 Tingkat kematangan gonad (TKG) yang didapatkan selama penelitian paling banyak dijumpai pada Fase TKG I dengan kisaran Indeks Kematangan Gonad (IKG) 0,04-17,9%, dan Fekunditas Ikan Sembilang bisa mencapai 690-2514 butir telur. Ukuran diameter telur ikan yang berbeda-beda dengan kisaran 3,01-4,12 mm dan pola pemijahan dikategorikan secara bertahap. Maka dapat disimpulkan bahwa Ikan Sembilang di didapatkan cenderung berada di TKG I dengan Rasio Kelamin 1;2.3 dan dikategorikan tidak seimbang. Fekunditas yang dihasilkan berkisar 690-2514 dengan pola pemijahan parsial atau terus menerus. Saran dari penelitian aspek biologi reproduksi ikan sembilang dilakukan setiap bulan untuk mengetahui TKG, dan Fekunditas serta ukuran diameter telur agar diketahui kapan pemijahan ikan akan terjadi, sehingga dapat melengkapi informasi yang sudah ada.
